Comment:
Uracil is converted to UMP primarily by uracil phosphoribosyltransferase, (UPRTase). In E.coli UPRTase catalyzes the Mg(2+) conversion of uracil and PRPP to UMP and PPi. The homotrimeric enzyme in E.coli is activated by GTP. In E.coli upp is the first of a bicistronic operon in which the second gene, uraA, encodes a cytoplasmic membrane protein required for uracil uptake. However, uraA does not seem to be contained in the U.urealyticum genome. Also, in E.coli the PRPP binding site is located at residues 123-138. In U.urealyticum, 13 of those 16 amino acid residues are either similar or identical.

The enzymatic pathway is:
UMP + pyrophosphate = uracil + 5-phospho-alpha-D-ribose 1-diphosphate.

Blast Summary:  PSI-Blast Search
Gapped BLAST revealed a number of significant hits against uracil phosphoribosyltransferase proteins. For example, residues 4-207 were 58% similar to the uracil phosphoribosyltransferase of B.caldolyticus (pid:e258331). Residues 3-207 were 54% similar to the probable uracil phosphoribosyltransferase of S.salivarius (UPP_STRSL), and residues 3-207 were 55% similar to the uracil phosphoribosyltransferase of L.lactis (UPP_LACLA).

Other significant hits were made to the uracil phosphoribosyltransferase proteins of B.subtilis, M.genitalium, M.hominis, M.pneumoniae, E.coli and H.influenzae.

A BLAST against the STDPRO database revealed significant hits against the upp of M.genitalium (MG030), where residues 3-207 are 51% similar, and a gene of T.pallidum (TP0448), a possible uracil phosphoribosyltransferase, where residues 6-205 are 24% similar.
