Essential step for de novo glycine and purine synthesis, DNA precursor synthesis, and for the conversion of dUMP to dTMP.

Dihydrofolate reductases are enzymes which catalyze the reduction of folic acid into tetrahydrofolic acid. They can be inhibited by a number of antagonists, such as trimethoprim (however trimethoprim resistant versions of this protein have been found in S.pneumoniae).

Blast Summary:  PSI-Blast Search
Gapped BLAST revealed a number of significant hits to the dihydrofolate reductase proteins of a number of other organisms. For example, residues 1-155 are 37% similar to DYR_MYCGE, residues 5-154 are 33% similar to a dihydrofolate reductase of S.pneumoniae (3513546). Residues 1-153 are 33% similar to a MG213 homolog in M.pneumoniae (Y213_MYCPN).

In addition to those hits, a number of significant hits were made to the dihydrofolate reductase proteins of M.pneumoniae (DYR_MYCPN), B.subtilis (DYR_BACSU), H.influenzae (DYR_HAEIN), L.lactis (DYR_LACLA), E.coli (DYR9_ECOLI) and S.aureus (DYRB_STAAU).

A BLAST in the STDPRO database revealed significant hits to the dihydrofolate reductase genes of both M.genitalium (MG228) and C.trachomatis (CT612), where residues 1-155 are 37% similar to MG228, and residues 19-156 are 26% similar to CT612.
